新型pH敏感性壳聚糖/明胶水凝胶的制备及其性能

摘要:用天然多糖壳聚糖和明胶制备了具有pH敏感性的壳聚糖/明胶水凝胶,研究了pH值对于该凝胶溶胀度的影响,采用红外光谱和电镜扫描对其结构进行了表征。结果表明,水凝胶在碱性和酸性环境中均具有PH敏感性,在酸性溶液中凝胶的溶胀比远大于碱性溶液中的溶胀比,其在pH值3.0时的溶胀度最大,在pH值9.0时的溶胀度最小,且其在不同pH溶液中重复可递溶胀收缩。同时,随着交联剂用量的增大,水凝胶的溶胀度减小。关健词:PH敏感性;壳聚糖;明胶;制激响应性

Abstract: Chitosan/gelatin hydrogels were prepared through the reaction between natural chitosan and gelatin. Swelling ratios of the hydrogels in buffer solutions at different pH values were studied. The structure was confirmed by FTIR and SEM. The results showed that the hydrogels were strongly pH-sensitive and the swelling ratio of the hydrogels in acid medium was higher than that in alkaline medium. Furthermore, the largest swelling ratio was obtained in a pH 3.0 buffer solution and the smallest was obtained in a pH 9.o buffer solution. The hydrogels had good swelling-contracting reversibility in the solution which pH value was alternately changed. With the increase of glutaraldehyde
dosage,the SR of the hydrogels decreased in buffer solutions at different pH values. Key words:pH-sensitive; chitosan; gelatin; stimulating response
智能水凝胶是对外界微细的物理或化学变化,如温度、PH值、离子强度、光和特异化学物质等做出响应的一类水凝胶,也被称为刺激响应性或敏感性凝胶(]。pH敏感性水凝胶是智能水凝胶的一大类,该类凝胶中含有大量的易水解或质子化的酸或碱性基团,可以根据环境pH值变化夺取或释放质子,使得水凝胶吸水溶胀或脱水收缩2)。壳聚糖(CTS)是含氮的多糖类物质,也是自然界唯一的天然阳离子多糖。近二十年来,随着对壳聚糖多种生物活性的发现,全世界对该类产品的研究十分活跃,其应用领域也不断拓宽。壳聚糖与人体内透明质酸和胶原的电荷性质类似,对保持蛋白质类药物或活性物质
的活性非常有利。由于水凝胶在结构与性能上与生物大分子存在许多相似性,这有助于模拟生物体内的过程、实现一些特殊的功能,因而其在生物医学领域有着巨大的应用前录,如膜、生物相容性材料、药物控释体系、药物和酶载体等[3-4]。明胶作为由动物胶原降解制得的生物蛋白大分子,其蛋白质结构决定了它有优异的生物功能,如生物降解性及良好的生物相容性,已广泛用于照相、食品及医药等工
