乙酸异戊酯高产菌脂肪酶的特性研究

摘要以乙酸异戊酯高产菌株Loq-c为试验材料从菌体中提取制备脂肪酶,研究其酶学特性结果表明该酶催化反应的最适温度在 30°C左右在40℃以下具有良好的热稳定性催化体系的pH在6.0~7.0时酶的相对活力和稳定性较高而当pH≤4.0时酶表现得不稳定,酶活力低。金属离子Mg"和Ca*能明显促进酶的催化能力Cu*和Ag*能明显抑制醇的催化能力Na、K"和Zn对酶催化的影响不显著。正交试验结果表明菌株Loq-c脂肪酶最佳酶活条件为温度在30℃pH值为7.0Mg浓度为5.0mmol/L。在此最佳条件下酶活力高达535.8U/mL

Characteristics of lipase from isoamyl acetate high-yield strain

Abstract : The lipase was extracted from isoamyl acetate high-yield strain Loq-c, and the enzyme characteristic was researched. Results indicated that e ass ee d o es e o e se , e e se ao range of 6.0-7.0, the relative activity and stability of lipase was both higher, but when the pH was less than or equal to 4.0, the relative activity and stability of lipase was both lower. The metal ion Mg* and Ca* could promote the enzyme catalytic ability significantly, and Cu' and Ag* could inhibit the enzyme catalytic ability significantly, but Na, K' and Zn* had no significant influence on enzyme catalytic. The orthogonal experiments results showed that the optimum reaction conditions of the lipase from strain Loq-c was temperature 30 °C, pH 7.0, Mg* 5.0 mmol/L. Under the optimum conditions, the enzyme activity was up to 535.8 U/ml

脂肪酶(EC3.1.1.3)普遍存在于动物、植物和微生物中[1-2]能催化三酰甘油酯与其他酯类物质的水解及合成反应此外还表现出一些其他酶的活性如酰肽水解酶、磷脂酶活性等14在粮油食品加工、日用化工、医药卫生、环境保护和能源开发等许多工业领域中得到广泛应用7]。
脂肪酶有多入同工酶其中种类最多的同工酶存在于
微生物中[78]如FICKERSP等[9]研究发现解脂耶氏酵母(Yarrowialipolytica)能编码合成16个脂肪酶同工酶。据统计产脂肪酶的微生物多达65个属其中脂肪酶生产能力较强的菌株主要有黑曲霉(Aspergillusniger)、根霉(Rhizopus Sp-)、粗状假丝醇母(Candidavalida)、假单胞菌(Pseudomonasaeruginosa)、链霉菌(Streptomyces)等(68)因此微生物脂肪酶是工业酶制剂的重要来源。菌株Loq-c是本实验室分离获得的一株高产乙酸异戊酯的克鲁维毕亦酵母(Pichiakluyveri)本研究初步从菌体中提取制备脂肪
