[Abstract] Objective: To investigate the integration of hepatitis B virus( HBV) DNA in peripheral blood mononu-clear cells(PBMCs) in the patients with hepatocellular carcinoma(HCC) , and to explore the mechanism of HBV -DNA in PBMCs in HCC patients. Methods: Fluorescent quantitative polymerase chain reaction(FQ PCR) was ap-plied to detect HBV - DNA both in serum and in PBMCs of 32 patients with HCC( HCC group) and 45 patients with chronic hepatitis B( CHB group) ,primers and probe were designed for HBV C gene. Results: HBV DNA positive rate in serum of HCC group and CHB group were 62. 5% (20/32) and 46. 7% (21/45) respectively, the average lev els of HBVDNA concentration were1o and 104copies/ml respectively;HBVDNA positiverates in PBMCs of HCC group and CHB group were 87. 5% (28/32) and 51. 1% (23/45) respectively ,the difference was sig nificant(P< 0. OI ), the average levels of HBV DNA concentration ( only HBV - DNA positive patients) were 10L3 and 10.5copies/ml respectively HBVDNA psitiverate in PBMCs of HCC group was higher than that in serum(87. 5% VS 62. 5% ,P <0. 05). HBV DNA of HCC patients and CHB patients were detected both in their serum and in PBMCs ,the HBV - DNA concentration in PBMCs were all lower than that in serum, the correlation of HBV DNA concentration were significantly positive(r =0. 9018, P <0. 01 in HCC group, and r = 0. 9025, P < O. O1 in CHB group,respectively). Conclusion:The integration of HBV DNA exists in PBMCs; by comparison with liver tissue,detecting HBV DNA concentration in PBMCs has the advantage with no harm as risky signal of predic ting HCC.

原发性肝细胞癌(PHCC)是全世界主要健康问题,在我国其死亡率已占恶性肿瘤死亡的第三位"]。研究显示HCC 发生与乙型肝炎病毒(HBV)的感染关系密切[2),HBV核酸(HBV-DNA)整合于宿主细胞是PHCC的发病机制之一[3]。
